Insulin Sensitivity and Beta-Cell Function Research demonstrated that GLP2 produces improvements in fundamental metabolic dysfunction: Approximately 8% reduction in HOMA2-IR (insulin resistance marker) with 10 mg dose Enhanced markers of beta-cell function including improved insulin secretory responses Greater insulin sensitivity improvements compared to selective GLP-1 receptor agonists Lower prandial insulin and glucagon concentrations compared to active comparators Obesity and Weight Management SURMOUNT Clinical Trials The SURMOUNT program investigated GLP2 for chronic weight management in participants with obesity or overweight: SURMOUNT-1 (adults without diabetes): Mean weight loss up to 20.9% (approximately 48 pounds) with 15 mg dose at 72 weeks compared to 3.1% with placebo SURMOUNT-2 (adults with type 2 diabetes): Mean weight loss up to 15.7% (34.4 pounds) with 15 mg dose at 72 weeks 55-63% of participants achieved at least 20% body weight reduction with higher doses Greater weight loss than achieved with selective GLP-1 receptor agonists in head-to-head comparisons Weight loss with GLP2 included favorable body composition changes, with fat-mass to lean-mass loss ratios similar to lifestyle interventions and bariatric surgery
